The movie tells the story of Ina Montecillo (played by Ai-ai delas Alas), a mother who has lost three husbands and is now raising twelve children by herself. Ina works many different jobs to take care of her big family, like construction work, selling pirated DVDs, and even being an impersonator in a nightclub. Even though she tries her best, Ina faces financial problems and disagreements with her kids, but she never gives up.

Each of Ina's children has different personalities and struggles. Juan, the eldest, takes a responsible role by helping her mother with family duties. Tudis, though a bit rebellious, deeply cares for her family despite arguments with her mother. Tri, the smart but rebellious son, while Por’s sweet nature makes her a helpful presence in the household. Pip brings laughter to the family and is more like a comedian, and Six, though prone to trouble, has a good heart. Seven stands out as an overachiever, and just like his older brother, Tri, she's also smart. Cate acts as a peacekeeper, while Shammy’s cheeky nature adds a playful element to family life. Ten-ten is a bit reserved, and the twins, although they are just babies, Connie and Sweet, bring joy to the family.
